旨在探讨拜城油鸡不同生长发育阶段小肠型脂肪酸结合蛋白(I-FABP)、载脂蛋白A1(APOA1)、微粒体甘油三酯转运蛋白(MTTP)、类微粒体甘油三酯转运蛋白(MTTPL)、脂肪分化相关蛋白(ADFP)和过氧化物酶体增殖物激活受体γ(PPARγ)基因与脂肪性状的关系,为深入研究候选基因对脂肪性状的作用奠定基础。采集拜城油鸡不同日龄(14、28、56、84、112 d)的胸肌、腿肌、肝脏、腹脂、背部脂肪和胸部脂肪样品共300份,测定其脂肪性状和血脂指标,采用实时荧光定量PCR技术对拜城油鸡不同生长阶段各组织中I-FABP、APOA1、MTTP、MTTPL、ADFP和PPARγ基因表达量进行检测。结果:I-FABP基因在14 d的肝脏中表达量最高,PPARγ基因在112 d的肝脏中表达量最高,APOA1、MTTPL基因在肝脏和肌肉中高表达,MTTP基因在腹脂中高表达,ADFP基因在胸部脂肪中高表达。相关性分析表明,I-FABP基因表达量与胸肌、腿肌的肌内脂肪(IMF)呈极显著正相关(P<0.01),与胸部脂肪重呈显著负相关(P<0.05),其在腹脂、背部脂肪和胸部脂肪中的表达量与低密度脂蛋白(LDL)呈显著正相关(P<0.05); MTTP基因在肝脏中的表达量与甘油三酯(TG)呈显著负相关(P<0.05),其在胸部脂肪中的表达量与LDL呈显著负相关(P<0.05); MTTPL基因表达量与腿肌IMF呈显著负相关(P<0.05),其在腹脂中的表达量与高密度脂蛋白(HDL)呈极显著正相关(P<0.01); PPARγ基因在背部脂肪中的表达量与LDL呈显著正相关(P<0.05)。结论:I-FABP、MTTP、MTTPL和PPARγ基因在拜城油鸡脂肪性状的形成过程中发挥重要作用。
Abstract:The aim of this study was to explore the relationship between intestinal the fatty acid binding protein( I-FABP),apolipoprotein A1( APOA1),microsomal triglyceride transfer protein( MTTP),microsomal triglyceride transfer protein-like( MTTPL),adipose differentiation-related protein( ADFP) and peroxisome proliferator-activated receptor γ( PPARγ) genes and the fat traits in Baicheng fatty chickens at different growth and development stages,and to lay a foundation for further study on the role of candidate genes in fat traits. A total of 300 samples of pectoral muscle,leg muscle,liver,abdominal fat,back fat and chest fat were collected from Baicheng fatty chickens at different ages( 14,28,56,84,112 days),and their fat traits and blood lipid indexes were determined. Real-time quantitative PCR was used to detect the expression levels of the I-FABP,APOA1,MTTP,MTTPL,ADFP and PPARγ genes in different tissues of the Baicheng fatty chickens at different growth stages. The results showed that the expression of the I-FABP gene was the highest in the liver at 14 days of age,the expression of the PPARγ gene was the highest in the liver at 112 days of age,the APOA1 and MTTPL genes were highly expressed in liver and muscle,the MTTP gene was highly expressed in abdominal fat,and the ADFP gene was highly expressed in chest fat. The correlation analysis showed that the expression of the I-FABP gene was significantly positively correlated with the intramuscular fat( IMF) of the pectoral muscle and leg muscle of the chickens( P<0. 01),and significantly negatively correlated with the chest fat weight( P<0. 05). The expression of the I-FABP gene in the abdominal fat,back fat and chest fat was significantly positively correlated with low density lipoprotein( LDL)( P<0. 05). The expression of the MTTP gene in liver was significantly negatively correlated with triglyceride( TG)( P<0. 05),and its expression in the chest fat was significantly negatively correlated with LDL( P<0. 05). The expression of the MTTPL gene was significantly negatively correlated with the leg muscle IMF( P< 0. 05),and its expression in the abdominal fat was significantly positively correlated with high density lipoprotein( HDL)( P< 0. 01). The expression of the PPARγ gene in the back fat was significantly positively correlated with LDL( P<0. 05). In conclusion,the I-FABP,MTTP,MTTPL and PPARγ genes played an important role in the formation of fat traits in Baicheng fatty chickens.
